Seal All is an excellent product if used right. Its not cheap, but it will work perfectly. Many shops that do extensive porting use it on everything they touch. NASCAR used it as well.

i was thinking the same thing. i wouldn't put it back together just to put a sealer in it. weld it now and forget about it.

Is that the kind that you pour in the radiator? I have the heads off right now.
Since you said the head is off.... have it welded

It seems that a couple of people are confused with the Seal All process. Its not something you pour into a engine that is already together. This is a sealer thats done with the head already off.Two totally different process'.Most often used to protect proting jobs and so forth, and it DOES work, IF done right. This isnt a can in the radiator fix.
I would tend to agree, if its off already, then weld it.
I answered the question that was asked. I was assuming there may be a reason the OP didnt want to, or possibly couldnt weld it. Dont know for sure.

Devcon or Hysol EA934, both 2 part epoxies with aluminum powder in the base. Both high-temp, aviation grade products. Once cured, they are harder than the aluminum.

Thats why I asked about alternatives to welding. The hole is in a very tough spot to get to , at least with the equipment that I have. It's right where the port floor starts to transition into the short side radius. I have everything here, that I would need , to try the seal-all. Cost is also a factor. The plant that I worked at closed last fall and I havent been able to find anything that comes close to what I was making before, so my race car budget is substantially less than it used to be.
