I kinda think you would be ok using some J B Weld stick, filling up that hole.
I too have one of those intakes, and when i got it, i put it in the bead cleaner to make it look spiffy.
The aluminum was porus on one runner, and started putting a hole in the top of the runner with the blast of the bead agent.
Used JB Weld on it, and it worked great, to this day.

I wouldn't think that small of a hole would have much if any effect on the fuel distribution. The DP4B's used to have a hole in the divider plate. I know because I ran a Holley three barrel on one back in the day. The rectangular hole or slot was necessary to clear the secondary throttle plate. It was not something I added, they were made that way.

The one I found 20yrs ago was cut also. That one from 20 years ago looks like it was already like that from the factory. I have a little Hot Rod Plymouth book that shows a factory drawing or blueprint and that looks correct. Maybe some were cut and some were't.

Yes, no doubt the plenum was cut, modified, for someones application. But they all started life as a 4 hole manifold, then after that, anybody could have done anything they wanted to, with the intake.
