There is a tool that can measure the volume of air that is drawn into the cylinder , not sure what it is called but they use it at some races to check engine cubic inches without taking the engine apart.

It's a P and G gauge. Below is some instructions on how they are used. Not your everyday, common tool though. Might be quicker to pull the pan and / or a head to check. PITA If it's running OK. https://www.precisionmeasure.com/product/cubic-inch-tester-500-670-800/

A stroker will be increasing the displacement through not so subtle extra stroke...so I'm thinking here that knowing the factory deck height, you might be able to literally put a metering stick of some sorts down the spark plug hole,and as perpendicular to the piston head as possible, and literally measure the distance it travels down into the bore as the piston goes to BDC.
Again, this is entirely relying on that "extra stroker" being around that 0.500" or greater measure....thus "not so subtle" as to be undetectable through such a simple measurement.
Crazy??? ...ummm, maybe, but if you have a compare baseline of some sorts you might be able to guesstime the number you are after!

For what effort you want to do, Cab's method (or some variation) is the best IMO.

A P&G gauge is over $1,000. I don't have one and I don't know anyone that does. Just thought this was funny...I remember hearing for some classes they were gonna tech cars w/ a P&G gauge. I've never seen a P&G gauge or saw a car getting checked w/ one, lol.

I think the pump the classes with cubic inch minimums even. There has been some small block cars win big block classes because drag week considers over 430" a big block unless it is a big block by architecture like a 383 chrysler or 396 chevrolet.

Round tracks used to use a P & G machine to "pump up" a engine. (it used air to measure the volume) If you know any round track guys local, you may ask them. Not sure if they're used any longer, with so many tracks using "crate" engines (which are sealed). Most just make you pull a cyl head if any question. But like others have said, it's just a number. What matters is how it runs.
Brian That's what they used at our local dirt track. My late machinist used to run in a 6 cylinder class back in the 70's. Engine of choice was 292 GM, 300 cube limit IIRC.They noticed when tech checked an engine, they only ever pumped #1 cylinder... Out came the engine, crank sent out to have journals 2 thru 6 offset ground to as much stroke as could be accommodated by whatever junkyard rods and shelf pistons were available. She was a winner! Do you have to know the exact Displacement or just 100% verify that it has been stroked?
Assuming the motor is out of the car and you can put the deck horizontal to the floor/tabe....just get a piston to BDC and pull a plug and keep putting a measurable volume of Marvel mystery oil (or some other light motor oil) compatible fluid into it. allowing some measurable displacement for the distance between the deck height, the head gacsket thickness and the approx distance between the head deck surface and the bottom of the spark plug hole, you can get pretty close. Of course your gonna be off by a bit for combustion chamber and piston eyebrows/dish/dome, but if you just want to do it quick and dirty that will work for a rough guess. A .030 over zero deck 440 would be about 913 ccs (just a little over a quart) plus whatever the make up distance (i mention above would be) is .
so a 4.15 stroke would already take over a litre (a couple ounces more than a quart) to fill just to the deck height. That (conveniently) seems pretty easy to determine.
To empty it, make some sort of nozzle that screws into the spark plug hole and run it through a hose and turn the crank.
It does't have to be accurate and engine is in the car
