Well with a Hemi block that kinda narrows it down. Predator and Hemi heads woudl be the only way to go.
As for the original question the B1 head has the best HP potential. If you can make 800+hp from a basic B1 combo you are doing something wrong. Getting to 800 from a -1 while not necessarily difficult will require a bit more thought.

I think there is a B1TS head available in a Hemi pattern, but that is an entirely different animal.

Yes, given what I already have, I was wondering how much power I could make on pump gas, CNC ported Stage V hemi heads, and a 4,5 bore megablock and 4,375 stroke (I think that comes up to 540). What would be the best combo of the other parts I need (intake, carbs, cam, etc.?). I already have 2.25" hemi headers for my A Body race car
Would a b1 setup have an edge over the above? Will it be cheaper to build? I can always trade my hemi megablock for a wedge....my headers would be useless though, Id have to fab new custom headers for my a body with stock inner fenders (arrghhh)

The 4.5 by 4.375 is 557(?) inches. A good build with 440-1 heads will make very good torque down low due to smaller ports than the B1. Up to about 800 hp with that many cubes port velocity should make the 440-1 a better choice for a reliable low rpm race motor. You should be able to shift at 6800 with a torqueflite and be in the sweet spot. The B1 will make more power but may need more rpm to do so. I shift my 528 with 440-1` heads at 7200, making 850+ hp.

I have had all three heads, Stage V on a pump gas blown hemi, CNC ported 440-1 on a pump gas motor and I do have a set of B1 not used yet. The B1 flow 430 cfm at .700, .800 and above, the 440-1 flow 370 at .700 and the ported Stage V flowed a tiny bit better than the B1s did If I was making the choice you have I would go with the Stage V based on the parts and intended usage nothing like the WOW factor of a Hemiroid

the best 440-1 flows 400 cfm and the best B1(not the TS) flows 470 cfm. Its true you have to use the B1 intake...but its about the best single four intake anyway. price wise they are about the same for the "package" ...Indy is about $3500 and you can get the B1 deal for around $3200 in the winter special. That said, if you want the biggest valve B1/MC it will cost another $900 for the Ti valves.
