the pins that the band levers ride on go in those holes..there is a threaded plug sealing the hole..looks like the rtv is insurance against a leak.

What worked for me on that issue was when I had the pan off was to clean the hole with brake kleen so the black RTV would adhere. Not sure if you need a regular O ring or a square cut one
It's a regular o ring , as said clean the hole good , put a big bead of RTV below the oring , install the tube and let the RTV cure overnight.
