Does the yellow fiber optic that attatches to #1 plug wire have anything to do with the start rertard?

Although I run the 7535 I thought the fiber optic was for a cam signal so the box knows where it is in the firing order.
I don't think it matters for the retard I think it's for individual cylinder timing.

go to MSDIGNITION.COM and look up the box number. it will tell you what each wire is for.

No, the start retard only works below 800 rpm ( or whatever you have it set to in Graphview). It doesn't use any wiring inputs to activate the start retard like the old AL3 needed.

should u be able to change timing in MSD an spin motor over, an see a difference with timing light?

In theory, yes. If you have the engine set at 30 deg, and have a 10 deg start retard, as long as its below 800 RPM your timing light should show 20. But I've honestly never tested it running. I usually use my MSD ignition tester with the laptop hooked up to make sure the retard numbers are activated correctly, which you can see with the "dials" function in Graphview. If you have an msd tester, hook it up and run it through the RPM range with the gauge function displayed from your box and you can watch the start ret and other stages come in and out. Or just hook up another distributor to the box and spin it by hand or drill.

Also just a tip if the box is new to you. Go into the run curve and drop the whole line down a few degrees, then set your timing manually where you want it. Then you can change timing up or down with a key stroke instead of wrenching it.
