Don't see why not with enough time, effort and planning.
The big stuff I can think of off the top of my head are;
Width - the Hemi is wider than a small block, so this will require doghouse modifications and keeping an eye on airflow, since all radiator and engine cooling air flows through here. If you keep the stock computer and thermostat, things could get hot in a bad way. Modern engines run at over 200, the old engine ran at 160-190. The new wiring should be able to handle it, but I doubt the doghouse insulation will.
Length - The fan to the bellhousing should work, but the transmission on the 100s was a shorty. The driveshaft was also short.
Are you also planning on brakes?
What about a suspension upgrade?
If you are doing a full-on update, you could go with an IRS and solve the trans/driveshart issue, since the diff stays in one place.

I like the points RodStRace raised, but want to stress out this particular one: WIDTH!!! You have to make sure that if you increase the doghouse width, you will still have room for the seats. The seats are right on the doghouse. Be sure to measure this before you commit. There's no real room to move the seats outwards. If it works, be sure to let us all know, the future is in the Gen III Hemis, but we want to know if they fit  Good luck, Ran

Some think it's backwards to carb a late model engine and ditch the EFI.I think it's more of an easy performance upgrade to the oe powerplant,be it a /6,or LA V8.You gain CID in this case anyway),better flowing heads,a roller cam ,a stronger v8 block,lighter weight,etc.EFI isn't always needed,especially on a fair weather limited use vehicle.In another thread moparts member Zippy mentions the possibility of an affordable distributor ignition conversion becoming available.Perhaps another step backwards,but it has it's right place in the right vehicle.Something like the A100 or an old school hot rod would be right with a distributor ignition.I've seen 426 and even 392 Hemis in A100's,the 3rd gen has to fit somehow!
