I have a factory inline street hemi intake manifold that's going on a mild stroker hemi...500+ cubes, 10:1 compression, mild roller, etc.
It's going to have a pair of Carter 9755S 750 cfm carbs on it. The car has 3.54 gears and is a 4 speed.

The manifold is unmolested. I haven't mocked it up yet, but it looks like I need to enlarge the front bores in the intake for these carbs to fit. Anything else I should do? Seems like the Vanke mod isn't a good idea for a street car...this car is 99% street driven. Any advice?

The Vanke mod would probably work fine in your application.

From Arruzza's website.

Street Hemi intake manifold . New from Mopar Performance . Every street Hemi intake I use on my engines or sell outright has the Arlen Vanke modification . My experience has proven this style intake MUST be modified in this way. The fuel distribution is far superior and the torque band starts sooner and finishes much later. There is no reason to run a stock manifold .

For many years I always heard that this modification hurts the bottom end, I never experianced that until I ran a set of 67 RO23 / WO23 750 cheater carbs , which is what Chrysler used on the “vanke” modifiefd intakes , Yes, they did lose bottom end, but that was the carbs , not the intake. If you have a crate hemi that needs a helping hand or are building a new engine , this Mopar intake with my machine work will deliver the goods

With 500 cubic inches I wouldn't think torque would be an issue so drive-ability should be fine.

That's what I was thinking too, but I've read a couple different things online saying it wasn't a good idea for a street car. I don't see it being a problem since this motor will have a 4.25" stroke in it (not sure on bore size yet). I'm asking here though b/c I have zero experience w/ this manifold and wanted to see if anyone has done something similar.

I have asked lots of people about it before, and on a STROKED HEMI, there is actually NO DOWNSIDE to doing it....just more power!!!! Just do it!

Back about 1990 I took off a stock Hemi intake and installed a Vanke modded intake on a "Fresh" stock 426 Hemi.
I noticed a small amount of bottom end torque difference but not so much that it was any problem with drive-ability.

I had the mod done on my intake on my 572 but I haven't fired it up yet much less driven it but I don't see street drive-ability as an issue.

No. But it fits stock aircleaners, hoods, Linkage, brackets etc.
