No, you cannot drive it without shifting.
Yes it will work with your stock shifter. There is a gate on your shifter that you must press the button in order to shift from 'D' to '2' , you can simply grind it off so that the shifter can be pulled from 'D' directly back to '2' without pressing the button , then you can press the button to shift from '2' into '3' . Remember it is reverse pattern, 'D' will be 1st gear , '2' will be 2nd gear and '1' will be 3rd gear.
With a manual valve body you will not need the kick-down linkage.

Skip the reverse manual trans unless you're building a drag car. There's no good reason for that on the street.

There's two types of "manual" valve bodies, the "manual/automatic" (B&M Transpak) where you retain your kick down linkage, and still retain "automatic" features, but when required you can have improved manual shifts, then there's the FULL manual valve body (B&M Manualpak), they can be built in "forward" (standard PNR321 pattern) or reverse pattern (PRN123), the kick down linkage is eliminated, full shifting is required all the time, just like driving a manual, but without a clutch pedal,...I had a reverse pattern full manual 904 then a full manual forward pattern 727 in my Challenger T/A before swapping in a 4 spd 883,....I rather enjoyed it on the strip or street, beaware there's other mods to perform to the tranny to accept a "manual valve body", in the way of changes to the govenor, and the servos, B&M makes a good "manualpak" kit for modifying your existing trans, attempt it only if your well versed in automatic transmission rebuilding, and have the tools to perform the work, as complete disassembly is required

Sure can , it will start in what ever gear you put it in and will never change from that gear .
The question I have is do you WANT a manual shift auto trans ?

Manual/automatic is a marketing gimmick....a bone stock VB can be shifted manually.

I have a reverses manual Valve body in my 69 Bee and love it. I installed a Hurst Pro-Matic under the counsel. I didn’t like using the stock shifter with the RMVB. It just didn’t feel right to me. I prefer the feel of the gates in the pro matic.
You can buy a new gate plate to change the stock shifter to correct the FEEL .

What is the feature that eliminates engine torque braking? Low band apply? You know- with a manual VB when you put it into 1st from 2nd the engine is decoupled from the trans- like putting a manual into neutral. I dont know the terminology but had 2 cars with full manual reverse VBs and both were no problems on the street.

Sorry Mike that B+M propaganda is a load of crap when it comes to a 727 , you can manually downshift to low above 15mph and it will not automaticlly upshift if not in Drive.
Plus the B+M shift improver KIT is a POS .

John, the Transpak, and Manual pak are not the "Shift Improver kit", I agree the "Shift Improver kit" is usless...
I generaly prefere the "manual pak", as it offers several build versions of Manual/Automatic features, as well as Full Manual control, I've built quite a few 904's and 727 for myself and customers over the last several decades, and had no issues with using the B&M Transpak, or Manual Pak

I've had a RMVB in my Charger for 14 years & I wouldnt have it any other way.
I use the stock console shifter but with a Duncan Motorsports RMVB gate that I was a test mule for.
Before the gate I would sometimes miss 2nd gear and go straight to third. Now with the gate the 1-2 shift is a "slap stick" style no button press. It has a positive stop on 3rd gear. 3rd requires a button push. It is also a slap stick style back up to first with a neutral lock out (must push button to get into neutral)
