i look at plugs and jet accordingly. if 2 and 4 look rich, i will put a leaner jet over them as an example. if thats what your asking

We used 8 egts on the dyno, and found the manifold combo needed 1 size larger on the left rear. It's been so many years, I forgot the numbers involved. I reckon each manifold would be different.

Its common to stagger jets to balance a manifold...
I use EGTs to do it but I change the manifold runners
to balance the manifold

On my 360 I saw a color pattern between cylinders being fed by one side of a Weiand Stealth dual plane and the other. The deeper plane was running leaner. I averaged out the numbers and had 2 or 3 jet sizes difference between the sides. The coloring evened out and I picked up about .03-.05.

Obviously doing it on a single plane would be a little more difficult to figure out. Other factors may also influence things. But basically on a single plane, all cylinders are being fed from a larger, single plenum so differences may not be so obvious without "tools".
