Yep you will need an adapter. Good news is Chrysler, Dodge and De Soto 1st Gen Hemis all use the same one and there are a few options. Is the 341 hemi any diff than the 354 and 392!? I was told by someone that the 318 poly trans will bolt up to a early hemi 341 Hemi is Desoto, 331/354/392 is Chrysler Hemi. Desoto had a 345 Hemi in the Adventurer.... same basic engine as a 341. 330 cubic inch Hemi too

Chrysler, Dodge and De Soto HEMIs each had unique blocks and heads with virtually no interchangeability between the brands.
The major thing that was standardized between the brands was the bellhousing and motor mount configuration. There were different amounts the crankshaft extended from the back of the block so in some applications you will find an aluminum spacer between the engine and bellhousing.
As far as using a 318 transmission it depends. The 57-61 cast iron case ones should be a bolt in. In 1962 Mopar introduced the aluminum case Torqueflite and at the same time shortened the amount of crankshaft overhang at the back of the block. The bellhousing bolt pattern was still virtually the same (the location of the lowest bellhousing bolt on the drivers side was moved inboard). Basically a 62 and up aluminum case Poly Torqueflite would physically bolt up to an early Hemi but without an adapter/spacer the longer crankshaft would cause the convertor to crush the front pump and/or crack the transmission case.
Additionally the first gen HEMIs used an 8 bolt crank flange drilled for 7/16" studs pressed into the converter/flywheel where the later 318 Cranks (I believe) are 6 bolt and and the crank flange is tapped for 1/2" bolts.
Basically the small block aluminum case torqueflite will require an adapter to use it behind an early HEMI.

For the OP's purpose, his DeSoto Hemi will take the same transmission adapter as all the pre-'62 Hemis and Polys except the '51-'54 Chrysler 331 with the extended block.

I'm currently doing an A-230 manual trans swap onto a 325 Poly in my '57 Dodge, I opted for the Hotheads adapter and flywheel. Nice product and the adapter puts the bell housing on an acceptable center (only .008 TIR). Why a three speed? Shifter on the column and you want to keep it?

Yup. Plus the A-230 has a better 1st gear for a heavy barge and synchro low. Had to do some creative machine work to get the shift levers to line up properly.
The bellhousing has both the A-230 and A-833 bolt patterns so an OD 833 is a possibility. (Retain the column shift and operate Reverse with a pull cable.)

Back to the original poster. Don't know what you are going to use for a convertor, but you will need to make sure that the one you use does not have a balance weight as the DeSoto Hemi is internally balanced.
