Looking at a 70 Cuda and inside the trunk is this capped off tube / filler neck on the passenger side of the trunk floor. Obviously this is not a fuel inlet given a Cuda's filler neck is centered behind the plate. Assuming it's a factory part, I'm interested to find out what this is / its purpose etc.

Probably part of the California Emissions system. There was a similar system for 1971 used in pretty much all cars, but it was changed/revised for '72.

Someone more knowledgeable than I will probably have all the info. I had a similar setup in a '71 Swinger. I think there were three hoses connected to the bottom.

I think you nailed it as this car was originally sold in California. Funny system, I'm sure it was very effective. Haha.

Yes it’s a clean air system. The fuel tank has four vent lines and it has a line going from the valve cover breather back to the rear to this tube. It might go to the tank. It’s been awhile since I looked at my cars CAS.

It's part of the ECS on California built or sold cars. My 70 TA also has the ECS. There should be 4 vent lines from the gas tank to the base of the unit. There should also be a hard line to the engine compartment that will attach to the breather with a rubber hose.
