I've been trying to think of potentials. You might pull the valve cover(s) & use your stethoscope.

Pull the distributor and see what the drive and the cam look like.

Best thing to do after an engine has been sitting up a long time is pull the distributer and prime to oiling system. I hope it's not a cam bearing issue. I had a faint whining noise once in one of my 440's that started never really seemed to effect anything. The one day it started running rough like it was on 6 cylinders. I had to drive it the 20 or so mile home. It was on 6 cylinders, spun a cam bearing & broke the cam shaft on the #4 journal. Cylinders #7 & #8 were not moving. The front 1-6 still ran fine & got me home. LOL. Cut the oil filter open & was filled of bearing material. I had to rebuild it & still going strong today.

Well....you might try some engine flush? Maybe some "smutz" in an oil passage? If you use that stuff, follow directions EXACTLY and only idle the motor for maybe 5 min. & replace oil/filter.
