I put a B-body rear in my Challenger. Will a 10" wide wheel that has a 4.5" backspace with a N50-15 tire fit?

Typically with a early b-body rear you need 5" BS. The later rears are a little wider and need 5.5" BS.

I have a 1970 b-body Dana in my Challenger with 9.5" wheel and 5.25 BS.

You should clarify which year B-body:
62-67
68-70
71-72

What is the location of the springs (i.e stock b-body perch or further off-set?)

It's a 69 rear with 1" offset hangers with the stock B body location perches.

For maximum tire fitment you need a 10" wheel with 5.25" backspacing.
